The loxP cassette present in TI{TI}wamKO-mc has been excised, removing the Disc\RFPmCh.Ubi marker. This leaves a single loxP site inserted in place of the endogenous wam locus (2.1kb of genome sequence is deleted, removing the entire wam transcription unit).
In wamKO homozygotes, mature sperm exhibit loss of flagellar motility and fail to fully individualize; seminal vesicles are devoid of sperm; sperm vesicle area progressively reduces in size; basal end of the testes exhibit an enlargement; individualization complexes appear abnormal; sperm flagella lack outer dynein arms on the microtubule doublets along the length of the axoneme; 64-cell cyst cell nuclei are reduced in size; mitochondria fail to properly align during meiosis; small clusters of unincorporated mitochondria are observed adjacent to the nebenkern; and mitochondrial derivatives are smaller when compared to control testes.
