A lexA tag has been inserted into the endogenous pyr locus, in between codons 456 and 457. In addition, a 1bp deletion occurred during the mutagenesis event, deleting the first C nucleotide of the codon encoding amino acid residue H295 (this event was likely due to faulty homology directed repair at the site of one of the gRNAs used). The frameshift results in an early termination codon after 28 frameshifted amino acids. Two separate translated products are produced from this mutated locus; an N-terminal fragment of pyr that includes the FGF domain and a C-terminal fragment of pyr that includes the transmembrane and intracellular domains and that is tagged with lexA. Expression of the C-terminal fragment is likely the result of internal initiation of translation from an in-frame start codon 3' of the introduced early termination codon.
