A sequence cassette consisting of 1. an attP site, and 2. a loxP cassette containing the Disc\RFPDsRed(T1).3xP3.cHa marker has been inserted into the endogenous Ipo9 locus, replacing almost all the transcription unit.
Embryos from Ipo9KO/Ipo9KO mothers show widespread mitotic defects during the earliest embryonic divisions (chromosome bridges, chromosome fragmentation, lack of chromosome condensation and an array of spindle defects).
Ipo9KO/Ipo9KO leads to meiotic defects during oogenesis: during stage 14 of oogenesis chromosomes are misoriented/mislocalized.
Ipo9KO/Ipo9KO leads to defects during spermatogenesis: chromosome segregation defects are apparent; the clustered post-meiotic nuclei remain round well beyond the stage when nuclear shape occurs; spermatids do not go through individualization (no actin cones or waste bags) and mature sperm is absent;at the onion stage there are variably sized nuclei and nebenkern.
