Description

Part of the endogenous Arp53D locus has been deleted and replaced with a sequence cassette that contains 1. an attP site and 2. a loxP cassette containing the Disc\RFPDsRed(T1).3xP3.cHa marker. The insertion has results in the introduction of an early stop codon in the Arp53D coding sequence.

Arp53DKO homo- and heterozygous males produce more adult progeny than 'wild-type' controls. Arp53DKO homozygous males produce more adult progeny than 'wild-type' controls in competition assays, but show lower fitness than 'wild-type' controls, as controls progressive increase in proportion of population over 20 generations.

Arp53DKO homozygous females produce fewer adult progeny at 29[o]C, but not at 25[o]C, whether crossed to Arp53DKO homo-, heterozygous, or 'wild-type' control males, although the phenotype is more severe when crossed to Arp53DKO homozygotes. According, Arp53DKO homozygous females produce fewer larvae at 29[o]C, whether crossed to Arp53DKO homozygous or 'wild-type' males, although the phenotype is more severe when crossed to Arp53DKO homozygotes.

Embryos from Arp53DKO homozygous parents exhibit abnormal nuclei that appear disorganized and lack compaction, as well as 'nuclear fallout', as compared to controls. Embryos from Arp53DKO/Arp53DWT.KI transheterozygotes also exhibit abnormal nuclei that appear disorganized and lack compaction, as well as 'nuclear fallout', as compared to controls.
