Adulthood-only, targeted CRISPR/Cas9-mediated knock-out of CapaR by expressing CapaRUAS.gRNAx4.pCFD6 and Spyo\Cas9UAS.P2 under the control of Scer\GAL4CapaR.1.5 and Scer\GAL4how-24B respectively (together with Gal80[ts] for the temporal control of expression), leads to decreased lifespan. In the Scer\GAL4how-24B knock-out condition, food consumption is decreased and locomotion is progressively decreased, as evidenced by the significant reductions in both morning and evening activity peaks; Upon rescue by the co-expression of CapaRUAS.cKa, locomotion is increased compared to controls.

Adulthood-only, targeted CRISPR/Cas9-mediated knock-out of CapaR, by expressing CapaRUAS.gRNAx4.pCFD6 and Spyo\Cas9UAS.P2 under the control of Scer\GAL4Act88F.1.3 (together with Gal80[ts] for the temporal control of expression), leads to a progressive increase in locomotion, as evidenced by significant increases in both morning and evening activity peaks.

Adulthood-only, targeted CRISPR/Cas9-mediated knock-out of CapaR, by expressing CapaRUAS.gRNAx4.pCFD6 and Spyo\Cas9UAS.P2 under the control of Scer\GAL4Akh.PL (together with Gal80[ts] for the temporal control of expression), leads to increased sensitivity (mortality) to starvation; the fat body exhibits lipid droplets that are fewer and smaller under starved conditions, and smaller under fed conditions, as compared to controls.
