The wGMR.PHb and Disc\RFPDsRed(T1).3xP3.cHa markers present downstream of the wild-type, GFP-tagged cno sequence in TI{TI}cnoGFP+ΔRA have been excised (using P1cre recombinase). The duplicated 3' portion of cno (encoding a truncated cno 'ΔRA' protein) that was present downstream of these two markers in TI{TI}cnoGFP+ΔRA has also been deleted (using homology-directed repair); this deleted sequence has been replaced by a loxP cassette containing a Disc\RFPDsRed(T1).3xP3.cHa marker. This results in a chromosome that expresses wild-type, GFP-tagged cno.
