The wGMR.PHb and Disc\RFPDsRed(T1).3xP3.cHa markers present downstream of the GFP-tagged 'ΔPDZ' cno sequence in TI{TI}cnoΔPDZ.GFP+ΔRA have been excised (using P1cre recombinase). The duplicated 3' portion of cno (encoding a truncated cno 'ΔRA' protein) that was present downstream of these two markers in TI{TI}cnoΔPDZ.GFP+ΔRA has also been deleted (using homology-directed repair); this deleted sequence has been replaced by a loxP cassette containing a Disc\RFPDsRed(T1).3xP3.cHa marker. This results in a chromosome that expresses GFP-tagged cno that lacks the PDZ domain (amino acid residues 1006-1099 are missing).
