Description

10xUAS regulatory sequences drive expression of codon-optimized shi coding sequence that also carries silent mutations that maintain the translated amino acid sequence but creates a sequence that is the target of the gRNA encoded by P{U6-shi.gRNA} (this gRNA does not target the endogenous shi gene). The gRNA target site is in the same region of the coding sequence as the lesion in shi1.
