Expression of IKKεNIG.2615R under the control of Scer\GAL4sca.PU results in abnormal bristle morphology, wherein bristles are short, branched and have a characteristic swollen region.
Bristles are short and branched in flies expressing ik2NIG.2615R under the control of Scer\GAL4ap-md544. Ultrastructurally, the actin bundles of the bristles are disorganised and some run through the cytoplasm. Actin bundle area and size are both significantly reduced compared to wild type, whereas the number of actin bundles is significantly increased. The number of microtubules is not significantly altered, and most retain the orientation parallel to the bristles' long axis. There is a significant increase in vesicle density and diameter within the bristles. Multivesicular bodies are rarely seen and there is a modest increase in mitochondria compared to wild type.
Bristles are shorter and thicker in the adult notum of Scer\GAL4sca-P309; ik2dsRNA.Scer\UAS flies.
The proximal dendrites remain intact for most ddaC neurons at 18 hours after puparium formation (APF) in animals expressing ik2dsRNA.Scer\UAS under the control of Scer\GAL4ppk.PG, indicating a defect in dendritic severing during dendrite pruning (proximal dendrites can be seen disconnected from the ddaC soma at 5 hours APF in wild-type animals).
The microtubules remain intact in the dendrites of ddaC neurons at 6 hours after puparium formation (APF) in animals expressing ik2dsRNA.Scer\UAS under the control of Scer\GAL4ppk.PG (microtubules in the dendrites of wild-type ddaC neurons show breakage at this stage).
Expression of ik2dsRNA.Scer\UAS in S2 cells, under the control of Scer\GAL4Act5C.PI, does not affect cell viability. However, these S2 cells show an increased likelihood to have a serrate or stellate morphology, instead of the typical round morphology.
Embryos treated with ik2 RNAi, generated by expression of ik2dsRNA.Scer\UAS under the control of Scer\GAL4btl.PS, show tracheal terminal branching defects. These include the duplication of terminal cells and the misorientation of the actin core.
Flies treated with ik2 RNAi, generated by expression of ik2dsRNA.Scer\UAS under the control of Scer\GAL4ap-md544, show gnarled, forked macrochaetae.
