Ends-out gene targeting has been used to introduce a mutation that changes the catalytic glutamic acid required for RNA slicer activity into an alanine into the endogenous aub locus on the SD-LosArenos chromosome. A P1\loxP cassette containing a w^+* marker has also been introduced downstream of the 3' end of the aub gene.
