A CCAP promoter regulates expression of a GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain).
An elav promoter regulates expression of a VP16(AD)::Zip+ hemidriver (component of a 'split driver' system, contains the VP16 activation domain). The hemidriver sequence includes the Tag:NLS(SV40-largeT) nuclear localization signal. An internal ribosomal entry site (IRES) from the Ubx gene followed by DsRed(Unk) coding sequence is present downstream of the hemidriver sequence, but this is not expressed in the nervous system.
ScerGAL4DBD.CCAP drives expression in larval CCAP neurons in the larval central nervous system, recapitulating the CCAP expression pattern, when combined with ScerGAL4AD.elav or HsimVP16AD.elav in a split-GAL4 system.
