The cytological location of dodeca was determined to be in the centromeric heterochromatin of 3R by in situ hybridisation to mitotic chromosomes using C(2)EN, T(Y;3)B197 and TM6B, compound and free chromosomes to cytologically distinguish autosomes and the arms of the third chromosome. In diploid mitotic cells dodeca sequences are found as one or two dots throughout the cell cycle. In interphase cells, hypotonic shock promotes decondensation of the genomic region containing dodeca. Colchicine treatment precociously separates sister chromatids and reveals dodeca within the intervening space connecting the main dodeca signals of each chromatid. Distribution of dodeca satellite sequence within a species is constant, but varies considerably between closely related species of the melanogaster subgroup, with D.mauritiana having dodeca sequences on the X, second and third chromosomes and D.yakuba and D.teissieri lacking detectable dodeca sequences.
An 11bp tandemly repeated sequence, Dmsc1, is a member of the dodeca satellite family.
Repetitive sequence dodeca satellite is present in D.melanogaster, D.simulans, D.buzzatii, D.hydei, D.immigrans and D.virilis. A related sequence is present in one plant and three mammalian species. dodeca satellite DNA maps to the centromeric regions of an autosome.
