The Mst77Y region was re-evaluated using long-read single molecule sequencing technology. It was found that 6 previously identified genes (Mst77Y-2, Mst77Y-8, Mst-77Y-9, Mst77Y-5Ψ, Mst77Y-11Ψ, and Mst77y-14Ψ) were probable artifacts. Multiple new genes were identified that correspond to duplicate copies of previously identified genes. The current release 6 assembly is not entirely correct, in the order or precise sequence of the genes. The current prediction is of 10 functional copies and 8 pseudogenes in the following order, all transcribed from the plus strand: Mst77Y-17Ψ, Mst77Y-10Ψ, Mst77Y-1, Mst77Y-13, Mst77Y-3Ψ, Mst77Y-7a, Mst77Y-12a, Mst77Y-7b, Mst77Y-6aΨ, Mst77Y-6bΨ, Mst77Y-12b, Mst77Y-4a, Mst77Y-12c, Mst77Y-4b, Mst77Y-4c, Mst77Y-15Ψ, Mst77Y-16Ψ, Mst77Y-18Ψ.
"Mst77Y-16Ψ" is one of 18 Y-linked genes that are related to the Mst77F gene (which is located on 3L). Evidence suggests that these multiple copies originated from a single duplication from the autosome to the Y chromosome, followed by duplications inside the Y. Ten of these Y-linked genes encode defective proteins (Mst77Y-3Ψ, Mst77Y-5Ψ, Mst77Y-6Ψ, Mst77Y-10Ψ, Mst77Y-11Ψ, Mst77Y-14Ψ, Mst77Y-15Ψ, Mst77Y-16Ψ, Mst77Y-17Ψ and Mst77Y-18Ψ) and the other eight are potentially functional (Mst77Y-1, Mst77Y-2, Mst77Y-4, Mst77Y-7, Mst77Y-8, Mst77Y-9, Mst77Y-12 and Mst77Y-13). The eight potentially functional genes produce approximately 20% of the functional Mst77F-like mRNA and evolved under purifying selection.
CR45779 may correspond to a redundant copy of Mst77Y-16Ψ (representing the probable true location on the Y chromosome, but less complete sequence).
