Interaction in vitro; bait present on live, transfected COS-7 cells; prey produced from conditioned media of transfected COS-7 cell line.

The interaction of Dscam with Netrin molecules is also observed for human and chicken orthologs. Chicken Netrin exhibits a similar affinity for the human Dscam and rat DCC proteins.

Binding was determined using a cell overlay assay. Dscam1 was expressed in COS-7 cells, and mixed with conditioned medium from cells expressing NetB. After a 45 minute incubation, cells were fixed and binding of NetB to the cell surface was detected by immunofluorescence using anti-Myc antibody.
