Co-transfection of Hrs with smo leads to increased levels of ubiquitinated smo.

Treatment of cells with hh-containing medium diminishes the Hrs-smo interaction and reduces the level of smo ubiquitination.

The interaction of Hrs with smo is not affected by overexpression/knockdown of Ubpy (a smo deubiquitinase), or by mutation of smo ubiquitination target sites.

The interaction of Hrs with smo is blocked by a phosphatase inhibitor, okadaic acid, and phosphomimetic smo mutations.
