Diagonal gels were performed by running a non-reducing dimension in a tube gel and a reducing second dimension in a slab gel. Under non-reducing conditions the disulfide bridge containing complexes appear delayed, but after the reducing second dimension these proteins will appear off the theoretical diagonal, given by a straight line from the origin of the gel and the signal from the monomeric forms (GstS1 and Tm1). The MAC141 antibody clearly detected heterodimers of Tm1 PK or Tm1 PF with Tm2.
