A set of third chromosome mutant stocks derived by insertional mutagenesis using the P-element construct P{lacW}; most lines have a lethal or semi-lethal phenotype. The P{lacW} construct carries a w+mC mini-white visible marker, Ecol\lacZ enhancer trap sequences, and bacterial sequences that allow plasmid rescue (FBrf0049800).
A genetic screen allowing selection of new insertions on the autosomes was carried out. Mobilization of the P{lacW} element occurred in males carrying a w[-] allele and P{lacW} on the X chromosome, and P{Δ2-3}99B, a stable source of P transposase, on a marked third chromosome. F1 males exhibiting the mini-white phenotype were recovered and a line for each fertile F1 male was established. Only lines with lethal or semi-lethal insertions mapping to the third chromosome were selected. To identify pre-meiotic clusters, complementation tests were carried out between lines derived from F1 males recovered from the same dysgenic cross; ~600 clusters were identified and eliminated from the collection.

2460 lines were characterized further, including determination of lethal phase. Using an array of characterized third chromosome deficiencies, 599 lines were mapped by failure to complement one or more deficiencies. The cytological locations of more than 1200 lines were determined by in situ hybridization to polytene chromosomes. For lines mapping to the cytological region 86E-87F, flanking regions were recovered by plasmid rescue and mapped by hybridization to genomic P1 clones from that region; for a limited number the flanking genomic sequence was determined.
