Cloning vector that can be used to generate a transgenic construct containing all three components of the flySAM (synergistic activation mediator) system, which is designed to produce effective transcriptional activation of a gene of interest. The vector contains 1. a single sgRNA scaffold into which two Tag:BS(MS2-CP) stem loops have been inserted (one on the tetraloop and one on stem loop 2) and which is expressed under the control of the constitutive ' U6:2 ' (snRNA:U6:96Ab) promoter, 2. a bicistronic transcription unit that is driven by 10XUAS regulatory sequences and encodes an artificial transcriptional activator containing a nuclease-dead form of Cas9 (dCas9::VP64), a self-cleaving T2A motif, and a transactivator protein (MCP::p65(h)::HSF1) which includes the Tag:MS2-CP tag. An sgRNA that targets sequence upstream of a gene of interest can be cloned into the sgRNA scaffold, and the attB site present can be used to integrate the sequence into an attP-containing docking site in the genome. The resulting transgenic construct contains the three components of the flySAM system, which when expressed together form a potent transcription activation complex that binds to the sgRNA target site, resulting in activation of the gene of interest.
