Use of the heat shock head involution defective (hid) conditional lethal
chromosomes. The heat shock hid transgenes were generated by Meagen Grether
in Herman Steller's lab (Genes Dev. 1995 9;1694). They were first adopted
for use in genetic screens by Lisa Moore in Ruth Lehmann's lab, and were
jumped onto balancer chromosomes by Mark Van Doren, also in Ruth's lab:
CyO, P{w+; HShid4}
TM3, Sb, P{w+; HShid14}
In order to eliminate heat shock hid chromosome containing offspring, we
have successfully used the following protocol:
\-let parents lay for 5 days in vials at room temp or 25C
\-remove parents
\-heat shock for 2 hrs. at 37C in a water bath on day 5
\-repeat heat shock on day 6
Notes:
Heat shocks must be done in a water bath. We have had no success with
using air incubators. Large water baths can be made from virtually any
large container along with a mountable heating circulation pump.
We have done the heat shocks in plastic vials. Using glass or using
bottles may require a slight modification of the heat shock protocol.
We have not extensively altered heat shock conditions. For the
particular insertion line and genetic background originally tested, heat
shock on two successive days reduced the number of escapers compared to
single heat shock. Other insertion lines and genetic backgrounds have
proved even more effective, and it is possible that a single heat shock
might be sufficient in some cases.
