Subject: Wnt2RJ and Wnt2I
The following information about the temperature sensitivity of Wnt2RJ and
WntI is quoted from a letter to the Bloomington Stock Center from Karen
Kozopas (3/03).
The ts nature of Wnt2(RJ) is described in Dev Biol. 2002 Mar
15;243(2):312-25. The ts nature of Wnt2(I) has not been published. The ts
phenotype is different for each allele, affecting the two tissues where
I've studied the phenotype differently:
This is the Wnt2 null phenotype in the two tissues:
Testis phenotype: loss of pigment cells, malformed or absent muscle layer
Held-out wing phenotype: wings held out at 90 degrees from body, loss or
misattachment of direct flight muscle (DFM) 52, misattachment of other DFMs
Allele Wnt2(RJ)/Df(2R)11 phenotype:
<up>note that the Wnt2(RJ) alllele has an associated lethal that I haven't
been able to get rid of by recombination, so I observed the ts phenotype in
the transheterozygotes</up>
Permissive temperature (18 degrees)
Testis phenotype: wild type
Held out wing phenotype: wild type
Nonpermissive temperature (29 degrees)
Testis phenotype: hypomorphic, loss of some pigment cells but little or no
muscle loss
Held out wing phenotype: wings held out at 45 to 90 degrees, loss of DFM
52, hypomorphic for misattachment of other DFMs
Allele Wnt2(I)
Permissive temperature (18 degrees)
Testis phenotype: wild type
Held out wing phenotype: wings are held out, DFMs not studied in detail
Nonpermissive temperature (29 degrees)
Testis phenotype: severe loss of pigment cells and muscle,
indistinguishable from null phenotype
Held out wing phenotype: wings are held out, DFMs not studied in detail
