WNT-TCF SIGNALING PATHWAY CORE COMPONENTS -
The canonical Wnt signaling pathway is initiated by the binding of a Wnt ligand to a frizzled family receptor on the cell surface. Activation of the pathway leads to the inhibition of cytoplasmic β-catenin (arm) degradation and its subsequent accumulation in the nucleus, where it regulates the transcription of target genes. Wnt2 transcript is first detected in stage 13 embryos, in the mesodermal cells which will form the somatic mesoderm of the gonad. Wnt2 is expressed in the gonad at stage 15, and this expression is limited to the posterior portion of the gonad at stage 16. In late third instar larvae, Wnt2 expression is detected in male genital primordia. In the 24 hr male pupal genital disc, expression is detected in the epithelial cells at the apical tip of the developing seminal vesicle. Wnt2 transcript is not detected in female structures; however, when Wnt2 is ectopically expressed in third instar larval females, male-specific pigment cells are formed in the resulting adults.

            Wnt2 promotes correct patterning of direct flight muscles through a mechanism that is independent of the attachment site differentiation initiated by sr.

            Wnt2 assists wg to specify the main tracheal trunk. Wnt2 affects tracheal development but apperently has no effect on the cuticle, whereas wg influences both.

            Candidate gene for testis length quantitative trait locus.

            Wnt2 is required for cell fate determination and morphogenesis in the developing male reproductive tract.

            Wnt2 encodes an amino terminal signal peptide suggesting the gene product is a secreted protein.
