Interaction in vitro; bait produced from transfected construct and present on live, transfected S2 cells; prey produced from conditioned media of transfected S2 cell line.

Wnt2 binds to the surface of S2 cells expressing otk or otk2, but not shg (DE-Cadherin).

GFP-tagged otk2 was transfected into S2 cells (which lack fz and fz2). Live cells were then mixed with Wnt2-containing conditioned medium at 4oC for three hours. Cells were then fixed and binding of Wnt2 to cell surface was detected by immunofluorescence using anti-Myc antibody.
