(From FlyBase curator) I am a curator for FlyBase, and am curating the genomic coordinates of several mutants described in Huynh, et al., 2004 (Developmental Cell 6:625-635 ). In particular, the mutants linha7E7-18 (Hrb27C7E7-18) and linha10B2-9 (Hrb27C10B2-9) are described as G to A transitions that change the tryptophans (W) at positions 312 and 342 to asparagines (N). However, any G to A transition in the Trp codon (TGG) must result in a stop codon (TAG or TGA); mutation to Asn requires that all three positions of the Trp codon be changed (TGG to AAT/C). Would it be possible for you to clarify this issue? (reply from D St Johnston) We have now re-sequenced both (hrp48 alleles) and it turns out that you are correct, i.e. both alleles are premature termination codons (TGG -> TGA), not substitutions with Asn.
