P{UAS-Chc.DN} expresses a dominant negative Chc protein under UAS control consisting of carboxy-terminal amino acids 1075 to 1678. BglII and KpnI sites and a start codon were introduced to flank the coding sequence via PCR amplification using the following oligonucleotides (Chc sequences are indicated in uppercase): 5'-ggagatctcaac atgAAGTTCGATGTGAAC-3' (5' oligo) and 5'-gcggtaccTCACAAGTAGGGATAGCCCAT-3' (3' oligo). The amplified fragment was subcloned into the BglII and KpnI sites of pP{UAST}.
P{UAS-Chc.DN}2 is a second chromosome insertion.
P{UAS-Chc.DN}3 is a third chromosome insertion.
