Microarray analysis of Df(2L)ast2
Vinayak Nikam, Chun L. Ng, Thomas Xu, Nicholas Schwartz, Diana Norton and Eric Spana
DNA samples from Df(2L)ast2 (FBab0001903) heterozygotes were compared to samples from wild type flies by Array Comparative Genomic Hybridization by the Duke University Biology 271L class (Spring 2011) as described in Erickson and Spana, 2006 (http://flybase.org/reports/FBrf0193934.html ). Corning CGAP slides spotted with the AROS Drosophila V1.1.1 ~70 nucleotide oligo set from Eurofins MWG Operon (www.operon.com) were used for the analysis, with most annotated genes represented by a single oligo denoted by a DM number. Sequences present at one copy in deletion heterozygotes were detected by lower relative fluorescence when compared to sequences present in two copies in wild type flies.
The left Df(2L)ast2 breakpoint lies within CG2839 or CG17941 or in the region between them, and lies in the range 2L:626409..640599 (R5) (Predicted cytology: 21E2) based on the following evidence.
The gene order at the left Df(2L)ast2 end is CG2826 (FBgn0040107), CG2839 (FBgn0031273), CG17941 (FBgn0000497). A sequence within CG17941 (DM00004954, 2L:640531..640599 (R5)) was deleted. A sequence within CG2839 (DM00006385, 2L:626341..626409 (R5)) was not deleted, but CG2839 may have been disrupted. A sequence within CG2826 (DM00006380, 2L:624740..624808 ) was not deleted.
The right Df(2L)ast2 breakpoint lies within CG31662 or CG31933 or in the region between them, and lies in the range 2L:1794258..1821591 (R5) (Predicted cytology: 22B2-22B3) based on the following evidence.
The gene order at the right Df(2L)ast2 end is CG31662 (FBgn0045501), CG31933 (FBgn0051933), CG31664 (FBgn0051664). A sequence within CG31662 (DM00015199, 2L:1794258..1794326 (R5)) was deleted. A sequence within CG31933 (DM00003918, 2L:1821591..1821659 (R5)) was not deleted but CG31933 may have been disrupted. A sequence within CG31664 ( 2L:1824404..1824472 (R5)) was not deleted.
The rest of the microarray data are consistent with genes between CG17941 and CG31662 (inclusive) being deleted in Df(2L)ast2.
