Microarray analysis of Df(2L)r10 Harrison Friedman, Insun Chong, Ben Chlebina, Danny Mammo, Diana Norton and Eric Spana DNA samples from Df(2L)r10 (FBab0001903) heterozygotes were compared to samples from wild type flies by Array Comparative Genomic Hybridization by the Duke University Biology 271L class (Spring 2011) as described in Erickson and Spana, 2006 (http://flybase.org/reports/FBrf0193934.html ). Corning CGAP slides spotted with the AROS Drosophila V1.1.1 ~70 nucleotide oligo set from Eurofins MWG Operon (www.operon.com) were used for the analysis, with most annotated genes represented by a single oligo denoted by a DM number. Sequences present at one copy in deletion heterozygotes were detected by lower relative fluorescence when compared to sequences present in two copies in wild type flies. The left Df(2L)r10 breakpoint lies within genes CG4161 and CG3956 or in the region between them, in the range 2L:15434573 .. 15476957 (R5) (Predicted cytology: 35D2) based on the following evidence. The gene order in this region of Df(2L)r10 is: CG4158 (FBgn0001983), CG4161 (FBgn0028892), CG3956 (FBgn0003448). A sequence within CG3956 (DM00007187, 2L:15476889..15476957 (R5)) was deleted. A sequence in CG4161 (DM00007336, 2L:15434505..15434573 (R5)) was not deleted, but CG4161 may have been disrupted. A sequence within CG4158 (DM00007334, 2L:15423627..15423695 (R5)) was not deleted. The right Df(2L)r10 breakpoint lies within CG5968 or CG42389 or in the region between them, in the range 2L:16549467..16559961 (R5) (Predicted cytology: 36A4-36A5) based on the following evidence. The gene order in this region of Df(2L)r10 is CG5968 (FBgn0032588), CG42389 (FBgn0259735), CG31815 (FBgn0051815). A sequence within CG5968 (DM00008767, 2L:16549467..16549535 ) was deleted, where a sequence from CG42389 (DM00001738, 2L:16559961..16560029 ) was not, but CG42389 may have been disrupted. A sequence within CG31815 (DM00015479, 2L:16607209..16607277 ) was not deleted. The rest of the microarray data are otherwise consistent with a deletion of the genes between and including CG3956 and CG5968.
