We recently noticed that the flybase information to the P-Element insertion line Dmel\P{lacW}l(3)s1754s1754 (FBti0005636) was updated and is available again, in Bloomington and the DGRC. We would like to suggest adding a comment on the lethality of this transposable element.
In our recent work we investigated the lethality associated to this stock and found evidence indicating that the lethality is not dependent on the insertion at position 15554702 on 3L, despite sitting at or in the first/second exon of CG7427. In previous experiments the insertion was found to complement the deficiency Df(3L)XG9 (/reports/FBrf0188732.html) and this information was used to estimate breakpoints of Df(3L)XG9. We continued investigation of Df(3L)XG9 and found that its breakpoints include position 15554702 (15524158-15659132). Amplification of genomic DNA of the CG7427 locus by PCR demonstrated that Df(3L)XG9 does uncover this gene. These findings indicate that the region causing the lethality on the chromosome carrying P{lacW}l(3)s1754s1754 must be located somewhere else on the chromosome.
In addition we found that the lethality of the P{lacW}l(3)s1754s1754 chromosome can be reverted to viability by delta2-3 mediated excision of the w*. Our suggestion is that the lethality is not coupled to P{lacW}l(3)s1754s1754 on this chromosome but potentially with another insertion on the 3rd chromosome. In addition based on the reversion results, we would also propose that PS1754 is not w* but a putative second transposon insertion is.
