PBac{U6-tracrRNA}VK00037 and M{U6-tracrRNA}ZH-2A are insertions of a donor attB construct into the PBac{y+-attP-3B}VK00037 and M{3xP3-RFP.attP}ZH-2A attP target sites, respectively. The sequence of the attB construct, named "pU6-tracrRNA", is available as the associated "pU6-tracrRNA.ape" file listed in the "Associated Information" section of this reference report. The construct contains the 89 nucleotide tracrRNA sequence from Streptococcus pyogenes described in Cong et al. (2013) ("Multiplex genome engineering using CRISPR/Cas systems" Science 339:819-23 ) driven by regulatory sequences for snRNA:U6:96Ab (FBgn0004189). It also contains a marker consisting of mini-white driven by GMR regulatory sequences (derived from the pGE-attB-GMR described in FBrf0208027 (Huang et al., 2009, Proc. Natl. Acad. Sci. U.S.A. 106(20): 8284--8289).
PBac{vas-Cas9}VK00027 and M{vas-Cas9}ZH-2A are insertions of a donor attB construct into the PBac{y+-attP-9A}VK00027 and M{3xP3-RFP.attP}ZH-2A attP target sites, respectively. The sequence of the attB construct, named "pvas-Cas9", is available as the associated "pvas-Cas9.ape" file listed in the "Associated Information" section of this reference report. "pvas-Cas9" is derived from the p3xP3-EGFP.vas-int.Dm.attB (FBtp0036670) vector and contains the Streptococcus pyogenes Cas9 gene sequence described in Cong et al., which has been tagged with nuclear localization sequences and codon optimized by mammals, driven by vasa (FBgn0262526) regulatory sequences. The construct also contains GFP expressed from the 3xP3 promoter as a transformation marker.
