Human PARIS, C571A and PARIS phosphomutant (DM) constructs were generated using standard cloning procedures by subcloning the respective cDNA sequence into pUAST plasmid (DGRC). Following sequence verification, constructs were microinjected into w1118 embryos (BestGene, Inc) and landed on the 2nd chromosome.
P{UAS-hZNF746.D}2 Human ZNF746 (aka PARIS) is expressed under UAS control. Insertion on chromosome 2.
P{UAS-hZNF746.C571A}2 Human ZNF746 (aka PARIS) with the C571A mutation is expressed under UAS control. The C571A mutation prevents ZNF746-dependent transcriptional repression of its target genes. Insertion on chromosome 2.
P{UAS-hZNF746.DM}2 Phospho mutant human ZNF746 (aka PARIS) is expressed under UAS control. Serine residues 322 and 613 were mutated to Alanine (double mutant, or 'DM') thus preventing phosphorylation and resulting in accumulation of toxic levels of ZNF746. Insertion on chromosome 2.
