P{10XQUAS(FRT.stop)TrpA1-T2A-mCD8GFP} consists of 10 copies of QUAS followed by a STOP cassette flanked by FRT, FRT3 and FRT5 sites. Downstream of the STOP cassette are coding sequences for TrpA1 followed by a T2A linker and mCD8GFP. Removal of the STOP cassette results in expression of TrpA1 and mCD8GFP under the control of QUAS. Full construct description: 10XQUAS-FRT-FRT3-FRT5-SV40 terminator-Tubulin alpha terminator-FRT-FRT3-FRT5-TrpA1-T2A-mCD8GFP-SV40 terminator. The two transcriptional terminators in the STOP cassette are the same as those used to make P{QUAS(FRT.stop)mCD8-GFP.P} in FBrf0210697.
P{10XQUAS(FRT.stop)TrpA1-T2A-mCD8GFP}2 is a viable insertion on the second chromosome.
P{10XQUAS(FRT.stop)TrpA1-T2A-mCD8GFP}3 is a homozygous viable and fertile insertion on the third chromosome.
