A transcriptional stop cassette that is flanked by three pairs of recombination sites (FRT, FRT3, FRT5) separates 10xQUAS from sequence that encodes two open reading frames (ORFs) separated by a T2A sequence (this allows each ORF to be translated separately). The two ORFs encode TrpA1 and Tag:M(mCd8a)-tagged GFP respectively. Excision of the stop cassette (using FLP recombinase) allows expression of the two ORFs.
