Tag:FKBP(F36V) is an artificial derivative of the Homo sapiens FKBP1A protein (HGNC:3711), carrying a F36V amino acid substitution. It can bind to a number of different synthetic ligands that have reduced affinity for the wild-type FKBP1A protein (so that addition of a synthetic ligand should not interfere with endogenous FKBP protein function) (PMID:10737745). Depending on the synthetic ligand used, binding to a ligand induces either homodimerization of Tag:FKBP(F36V), or induces heterodimerization of Tag:FKBP(F36V) with another conditional binding tool that is compatible with the synthetic ligand. These properties can be exploited to study the function and/or manipulate the activity of a tagged protein of interest, to bring together two tagged proteins of interest, or to target a protein of interest to a particular subcellular location (reviewed in PMID:29590011).
