Description

Fragment of Sry-β upstream region from -324 to +137 (where +1 refers to the Sry-β transcription start site) replaces the Sry-δ sequences. The first four amino acids of Sry-β are fused in frame to lacZ.

In EcollacZ-reporter constructs driven by either -324 to +137 or the -117 to +137 Sry-β sequence (+1 is the Sry-β transcription start site), expression is detected at high levels in the germarium and nurse cells from stage S1 to S7. Follicle cells begin to stain for EcollacZ at stage S7, and by stage S10, follicle cell staining is strong, while expression is faint in nurse cells and oocytes. In adult males, staining is detected in the male accessory gland, the ejaculatory duct, and in the non-apical portions of the testis (suggestinghat expression begins in late premeiosis). Staining is also seen in larvae, in larval tissue, the gonads of both sexes, and imaginal tissue.
