Transgenic construct generated in vivo by excision of the FRT cassette in P{Winkelried}. Contains the following elements: a scs element, the Ecol\lacZHsp70Bb.-190 reporter, a single FRT site, a w+* minigene that includes the eye-specific enhancer element, an scs' element. The presence of the flanking scs and scs' elements is designed to insulate the lacZ reporter and w+* minigene from euchromatic position effects, while they remain sensitive to heterochromatic position effect variegation (PEV).
Hsp70 promoter, 5' untranslated region (UTR) and first seven codons are fused to the lacZ coding region, Hsp70 3' UTR sequences are present.
Expression is fully heat-shock-inducible in virtually all cells during all stages, except pre-blastoderm embryos. In third instar larval salivary gland, detectable expression without heat shock.
