Description

Transgenic construct that is designed to test the effect in-cis of a protein on heterochromatic position effect variegation (PEV). The transgene contains the following elements: a scs element, the Ecol\lacZHsp70Bb.-190 reporter, an FRT cassette (containing three UAS sequences and four E2F binding sites), a w+* minigene that includes the eye-specific enhancer element, an scs' element. The presence of the flanking scs and scs' elements is designed to insulate the lacZ reporter and w+* minigene from euchromatic position effects, while they remain sensitive to heterochromatic PEV. If a protein of interest is fused to GAL4(DBD) it can bind the UAS sites present in the FRT cassette, and its effect on PEV of the lacZ reporter and w+* minigene can be analysed.
