The His3.3B promoter drives expression of the wild-type His3.3B coding sequence tagged at the C-terminal end with Tag:FLAG and followed by His3.3B 3' UTR sequences. The His3.3B coding sequence, Tag:FLAG tag and 3' UTR sequences are contained within an FRT cassette and can thus be removed by using FLP recombinase.
Promoterless mutated form of His3.3B (amino acid replacement K4A), which is tagged at the C-terminal end with Tag:HA and followed by His3.3B 3' UTR sequences. The FRT cassette which is present in P{His3.3B(FRT.His3.3B.WT.FLAG.ubiGFP)His3.3B.K4A.HA} prevents expression of this mutated form of His3.3B.
The P{His3.3B(FRT.His3.3B.WT.FLAG.ubiGFP)His3.3B.K4A.HA} construct consists of the His3.3B promoter, followed by an Scer\FRT cassette which contains the wild-type His3.3B coding sequence tagged at the C-terminal end with T:Zzzz\FLAG, His3.3B 3' UTR sequences and also contains a Avic\GFPE.Ubi marker. Following the Scer\FRT cassette is a promoterless mutated form of His3.3B (amino acid replacement K4A), which is tagged at the C-terminal end with T:Ivir\HA1 and followed by His3.3B 3' UTR sequences. When the Scer\FRT cassette is present, the wild-type His3.3B sequences are expressed under the control of the His3.3B promoter (represented by His3.3BT:Zzzz\FLAG) and the mutated promoterless His3.3B sequences (represented by His3.3BScer\FRT.K4A.T:Ivir\HA1) are not expressed.
