The P{10XUAS-flySAM1.0} construct encodes two components of the 'flySAM' (synergistic activation mediator) system, which is designed to produce effective transcriptional activation of a gene of interest. P{10XUAS-flySAM1.0} encodes the dCas9::VP64 and MCP::p65(m)::HSF1 proteins separated by a self-cleaving T2A motif, expressed under the control of 10XUAS regulatory sequences. The third component of the flySAM system is an sgRNA that is tagged with two Tag:BS(MS2-CP) stem loops (which are specifically bound by MCP::p65(m)::HSF1) and which targets sequence upstream of the gene of interest. When these three components are expressed together, they form a potent transcription activation complex that binds to the sgRNA target site, resulting in activation of the gene of interest.
