UASp regulatory sequences drive expression of polo that carries mutations in the polo-box domain (amino acid replacements W395F, H518A, K520A), in residues analogous to those required for phospho-binding in human Plk1. The coding sequence is tagged at the C-terminal end with GFP. Synonymous codon replacements have been introduced into the transgene, so that it is insensitive to RNAi mediated by P{GD7563}.
