P{UAS-Cerulean.intMEMOIR} is composed of Cerulean plus 10 unique barcodes, each one of which is flanked by unique attP(Bxb1)/attB(Bxb1) sites recognized by Bxb1:int integrase, all under the control of UAS. In the presence of Bxb1:int, integrase activity can result in no change or stochastic deletion or inversion of individual barcodes. Readout of each barcode status via sequential rounds of single molecule FISH allows the generation of cell lineage trees.
