Mi{hsGFP} represents a transgenic construct generated in vivo by using phiC31:int-mediated recombination to replace the attP cassette of a Mi{MIC} element insertion with a cassette containing the fluorescent Avic\GFPHsp70.PD marker (donor sequence containing this cassette can be released from the P{FRT(attB.hsGFP.attB)} donor construct by FLP-mediated excision). Mi{hsGFP} is designed as an in vivo reporter for Polycomb group mediated gene silencing. It can be used to study silencing in the female germline, since the short Hsp70 5'UTR sequence present in the Avic\GFPHsp70.PD marker is designed to render the reporter insensitive to ovarian Hsp70-directed piRNAs.
