Description

UASt regulatory sequences drive expression of an 'ER-phagy' sensor designed for analysis of the dynamics of endoplasmic reticulum (ER) autophagic flux. The sensor consists of a Tag:SS(BiP) signal sequence, the pHluorinSE (pH-sensitive) and mKate2 (pH-resistant) fluorescent proteins, a Tag:ER(KDEL) ER retention signal and the Tag:V5 epitope. Under normal conditions, fluorescence from both pHluorinSE and mKate2 occurs. Upon clearance of endoplasmic reticulum (ER) by 'ER-phagy', in which ER fragments are delivered to autophagosomes and then fuse with lysosomes, the low pH of the lysosome quenches the pHluorinSE signal but not the mKate2 signal.
