UASt regulatory sequences drive expression of an inverted repeat.
Flies expressing Hsp22GD8704 under the control of Scer\GAL4GMR.PU show no defects in ommatidial structure.
Short-term recovery from chilling at 0[o]C is significantly affected in Scer\GAL4Act5C.PI/Hsp22GD8704 flies. After 60 minutes, all Scer\GAL4Act5C.PI/+ flies recover, while 24% of Scer\GAL4Act5C.PI/Hsp22GD8704 flies remain in a chill coma. All flies eventually recover and no mortality is observed.
Medium-term chill-recovery tests reveal significant differences in mobility (climbing) between Scer\GAL4Act5C.PI/+ and Scer\GAL4Act5C.PI/Hsp22GD8704 flies. Differences are manifested after 2 hours and 4 hours of recovery, with, respectively, 74% and 40% f flies still injured in Hsp22GD8704-expressing flies with 52% and 15% in Scer\GAL4Act5C.PI heterozygotes.
Adults expressing Hsp22GD8704 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
