Homozygous U2af50XR15 mutants display a temperature-sensitive phenotype when grown at a restrictive temperature (30oC). The homozygous females lay eggs with abnormal eggshell morphology, and the resultant mutant embryos fail to develop into larvae.

Germline clone analysis demonstrates cell lethality.

U2af50+mBa efficiently rescues the U2af50XR15 deletion with a similar rescue percentage at both 25oC and 29oC. Crossing four transgenic lines containing U2af50D204N.ts with U2af50XR15 results in rescue of the U2af50XR15 phenotype in two of the lines (P{U2af50.D204N.ts}1 and P{U2af50.D204N.ts}4), with an optimal rescue of 39% at 25oC and 15% at 29oC in one of the lines (P{U2af50.D204N.ts}1). Crossing three transgenic lines containing U2af50S284Y.ts with U2af50XR15 results in rescue of the U2af50XR15 phenotype in two of the three lines (P{U2af50.S284Y.ts}1 and P{U2af50.S284Y.ts}2), with an optimal rescue of 96% at 25oC and 49% at 29oC in one of the lines (P{U2af50.S284Y.ts}2).

U2af38ΔE18 U2af50XR15 double mutants can be rescued by a combination of U2af50+tS12 and U2af38+t10 but not by a combination of U2af50ΔRS and U2af38ΔRS. Lethal combination could both be rescued by an identical synthetic RS domain by a combination of U2af50RS.7 and U2af38ΔRS.
