Expression of bcdprotein from a single bcd+t8.7 copy recapitulates the wild type bcdprotein expression pattern, demonstrating high bcd protein concentration atthe anterior tip of the embryo. Additional copies of bcd+t8.7 increase thelevel of bcd protein in the embryo, where high protein concentrations aredetected at up to 50% egg length with four copies.
Embryos which are carrying bcd+t8.7 and have six copies of bcd+ show a posterior shift of the pre-cephalic furrow domain (which has small yolk stalk diameters and a greater depth) during cellularisation. The anterior cellularisation domain of large yolk stalk diameters expands to cover close to half of the embryo. The anterior domain of lower nuclear density seen in wild-type embryos is expanded posteriorly.
Embryos carrying seven copies of bcd+ (five in constructs and two wild type genomic copies) show failure to regulate cell number. Embryos have an enlarged head and the parasegments of the trunk are crowded into a reduced space, 5% reduction in number of bristles in A3, 31% reduction in cell number in parasegment 8 and histoblasts of the abdomen show a 40% reduction. One quarter of embryos hatch to larvae, these show a severe disruption of denticle pattern. Adults have gross defects in abdominal segmentation.
