Description

Construct: Coding region of the first 90 amino acids from Src64B is fused in frame to the coding region for csw amino terminus. Mutant protein is expressed under the transcriptional control of an Hsp70 promoter and two copies of the sev enhancer.

Flies that express one copy of csw::Src64Bhs.2sev in developing eyes often display extra R7 photoreceptor cells; while over 30% of such flies show the wild-type number of one R7 cell per ommatidium, over 30% show 2 per ommatidium, ~20% show 3 per ommatidium and a small number of flies show 4, 5, or even 6 R7 cells.

Flies carrying csw::Src64Bhs.2sev have an average of 1.2 ectopic R7 cells per ommatidium.

Eyes of transformed flies are slightly roughened. Roughness is due to the recruitment of cone cell precursors to the R7 fate. R7 cells can even be induced in ommatidia that otherwise lack R7 function. Ras85DN17.hs.2sev completely blocks the ability of csw::Src64Bhs.2sev to induce R7 development.

Flies that express one copy of csw::Src64Bhs.2sev in developing eyes with a styΔ5/+ background show an enhancement of the extra R7 photoreceptor cell phenotype seen when csw::Src64Bhs.2sev is expressed in a wild-type background. A higher proportion of the csw::Src64Bhs.2sev/+; styΔ5/+ flies show 3, 4, 5,or 6 R7 photoreceptor cells per ommatidium.

mask10.22 dominantly suppresses the ectopic R7 phenotype caused by csw::Src64Bhs.2sev; the double mutant flies have an average of 0.6 ectopic R7 cells per ommatidium. mask5.8 dominantly suppresses the ectopic R7 phenotype caused by csw::Src64Bhs.2sev; the double mutant flies have an average of 0.8 ectopic R7 cells per ommatidium.

Rescues the zygotic lethality of csw13-87/Y males.
