The full length Mmus\Hoxa5 open reading frame is expressed under the control of an Hsp70 promoter.
Expression of Mmus\Hoxa5hs.PZ using a 50 minute heat shock at 5.5 hours of development results in 98% of first instar larvae having an ectopic T1 beard in thoracic segment T2 and/or T3.
Expression of Mmus\Hoxa5hs.PZ using a 1 hour heat shock at 5.5-6.5 hours of development results in Scr-like transformations. Characteristically, thoracic segments T2 and T3 are transformed towards T1; the ventral beard, normally only found in segment T1, is seen in the T2 and T3 segments. Failure of head involution is seen and many cuticular head structures, such as the epistomal sclerite, ectostomal and hypostomal sclerites, H-piece, mouth hooks and sensory organs, are absent or misplaced. No transformation of the cuticle pattern is detectable in the abdominal segments. Ectopic expression of Mmus\Hoxa5hs.PZ during the larval stages using multiple heat shocks produces Scr-like transformations in the external head structures of adult flies. The abnormalities include reduced and irregular compound eyes, malformation of the mouth parts, thickening of the arista, a Ctx-like transformation in the hindhead, splitting of the maxillary palps into two segments and transformation of the antenna to leg. Structures resembling sex comb teeth are occasionally seen on the transformed antennal legs.
