A deletion removes the majority of the N-terminal region of the H3 domain (amino acids 204-250).
In contrast with wild-type, Syx1AΔ229 and Syx1A06737 flies rescued with Syx1AH3-N, undergo no peristaltic muscle contractions prior to hatching, nor show any response to tactile stimulation. In Syx1AΔ229 embryos rescued with Syx1AH3-N (as in Syx1AΔ229 embryos) no denticle belts are seen. Using whole-cell patch-clamp analysis, in Syx1AH3-N mutants in a Syx1AΔ229 null background, evoked and spontaneous neurotransmission is abolished. Postsynaptic muscles respond normally to neurotransmitter.
