Expression of sytScer\UAS.cLa under the control of Scer\GAL44G results in a significant increase in FM1-43 dye uptake at the neuromuscular junction after stimulus compared to controls.
Overexpression of sytScer\UAS.cLa in neurons, driven by Scer\GAL4elav.PLu creates larvae that when tested electrophysiologically by stimulation of the neuromuscular junction, produce peak amplitudes of the excitatory junction potential that are not significantly different from wild-type. No decrease in muscle miniature excitatory junction potential (MEJP) amplitude or muscle resting membrane potential is seen in sytScer\UAS.cLa,Scer\GAL4elav.PLu larvae, however MEJP frequency is decreased by about a third.

The lethality of stnA1 stnB1/Y ("stn8P1") animals is not rescued by expression of sytScer\UAS.cLa under the control of either Scer\GAL4elav-C155 or Scer\GAL4shi.PS. The lethality of stnA12 stnB12/Y ("stn13-120") animals is not rescued by expression of sytScer\UAS.cLa under the control of either Scer\GAL4elav-C155 or Scer\GAL4shi.PS.
stnA12 stnB12 homozygotes show embryonic lethality. Expression of sytScer\UAS.cLa under the control of Scer\GAL41407 in this background results in approximately 98% of animals hatching at normal times, but they then die as first instar larvae. Expression of sytScer\UAS.cLa under the control of Scer\GAL44G in the stnA12 stnB12 background results in approximately 96% of embryos hatching, although hatching is delayed (animals hatch between 21 and 35 hours after fertilisation. These animals show adult viability. Expression of sytScer\UAS.cLa under the control of Scer\GAL44G rescues the endocytotic functional defects seen in stnA15 stnB15 ("stnC") mutants.
Expression of Syt1Scer\UAS.cLa under the control of Scer\GAL4elav.PLu completely rescues the defects in the evoked synaptic currents recorded from muscle 6 of Syt1N13/Syt1AD4 embryos following motor nerve stimulation in 4mM extracellular Ca[2+]; evoked release is fully synchronous, and few asynchronous release events are seen in the rescued animals.
Transgenic expression of sytScer\UAS.cLa (under the regulation of Scer\GAL4elav-C155) in the null sytAD4/sytN13 background is able to partially restore all the behavioral defects observed. Overexpression of sytScer\UAS.cLa (under the regulation of Scer\GAL4elav-C155) is able to restore evoked excitatory junctional potential (EJP) amplitudes to near wild-type levels in sytAD4/sytN13 mutants.
