A duplicated sev enhancer and Hsp70 promoter drive expression of a mutated dos cDNA; all Tyr residues outside the DOS PH domain except Y801 and Y854 have either been removed by deletion (in frame deletion of codons 211-507) or mutated to Phe.

Comments

The addition of dos^YY.sev.hs strongly suppresses the rough eye phenotype seen in csw^CS.hs.2sev; dos^1.46 flies. In dos^1.46/dos^2.42 flies rescued by dos^YY.sev.hs, occasional ommatidia are observed that lack a R7 or R1-6 photoreceptor cell. Heat shock expression of dos^YY.sev.hs rescues the lethality and almost rescues the eye phenotype seen in dos^1.46/dos^2.42 or homozygous dos^P115 flies.
