P{EP}IncenpEP2340 is inserted into the third exon of Incenp.
Male flies carrying IncenpEP2340 show a dominant meiotic phenotype that results in low fertility. The main defects incude misshapen meiotic spindles, tetrapolar or multipolar spindles and defects in onion stage spermatids consistent with problems both in chromosome disjunction and cytokinesis. Heterozygous individuals also show defects in larval mitosis and mild external defects consistent with abnormalities during imaginal mitosis, including slightly roughened eyes, nicked wings and defects in the abdominal terguites.
Heterozygotes show reduced fertility.
Mutant males show defects in meiosis I; cells are seen in which unaligned chromosomes are distributed along the spindle and cells in which the four bivalents are not distinguishable or the chromosome morphology is abnormal are seen.
Heterozygous males show increased nondisjunction of the sex, second and fourth chromosomes during meiosis compared to controls.
No oocytes have been observed in females in which homozygous IncenpEP2340 germline clones have been induced, suggesting that Incenp is required for early stages of oocyte development.
Expression of IncenpEP2340 under the control of Scer\GAL4ey.PH results in eyes that are strongly reduced in size. Flies expressing IncenpEP2340 under the control of Scer\GAL4hs.2sev have wild-type eyes. Flies expressing IncenpEP2340 under the control of Scer\GAL4en-e16E have wings with a smaller posterior compartment that contains a reduced number of cells. Cells from third larval instar wing discs expressing IncenpEP2340 under the control of Scer\GAL4Act5C.PP are normal in size. There is a small increase in the proportion of cells with a 4N DNA content (cells presumed to be in G2) compared to controls. Homozygous blastoderm embryos have irregular cell outlines after cellularisation and appear to contain increased amounts of DNA.
